F-boxタンパク質β-TrCPの生理的基質の検索と生体における機能解析

研究概要

タンパク質の発現量調節が個体の発生の関係を明らかとすることが本研究の目標である。F-boxタンパク質
β-TrCP2/Fbw1b(以下β-TrCP2)の機能解析を行い、個体の発生・分化における役割を検討した。
β-TrCP/Fbw1(以下β-TrCP)はβ-cateninやIkBαなどをユビキチン化するユビキチンリガーゼである。しかしながら、われわれが作製したβ-TrCP1ノックアウトマウスでは、β-cateninやIkBαの分解がなお観察されることから、β-cateninやIkBαの分解を担うユビキチンリガーゼはβ-TrCP1のみではなく、またβ-TrCP1の生理的基質は他の分子である可能性が残された。一方β-TrCP1の相同分子であるβ-TrCP2のノックアウトマウスは、胎生9.5日に胎生致死である。このことはβ-TrCP1とβ-TrCP2の間に生化学的に差違は認められていないにも関わらず、生物学的には大きな差を持っていることを示している。
そこでβ-TrCP2の認識配列を持つタンパク質の中で、胎生初期から中期にかけて発現しているものを4個ピックアップし、β-TrCP2との結合能やβ-TrCP2によるユビキチン化の有無を検討しが、いずれも結合を認めず、またβ-TrCP2による分解促進は観察されなかった。
一方、β-TrCP2のコンディショナルノックアウトマウスの作製を行った。
β-TrCP1のノックアウト胎仔線維芽細胞では、特にPer2の制御異常が観察され、概日リズムの維持にβ-TrCP1によるタンパク質分解が重要な機能を果たしていることが示唆された。
