研究概要

本研究では、ネクチンーアファディン系の細胞間接着による細胞極性の形成機構に焦点を絞り、ネクチンーアファディン系の細胞間接着の形成から接着シグナリングを介したタンパク質および細胞骨格の不均等な局在の形成機構、すなわち細胞極性確立の分子機構を明らかにすることを目的として、研究を行なった。研究の結果、次のような研究成果を得た。ネクチンによるアドヘレンスジャンクションおよびタイトジャンクションの形成には、アファディンとアクチン細胞骨格が必須の役割を果たすことを明らかにした。アネキシンIIをノックダウンした細胞では、タイトジャンクションはカドヘリンによる細胞間接着形成なしに形成されること、タイトジャンクションの形成にはネクチンーアファディン系は必須であることを明らかにした。さらに、アドヘレンスジャンクションおよびタイトジャンクションの形成には、アファディンとPar-3が協調的に機能することを明らかにした。
