研究概要

獲得免疫系の最大の特徴は、その主体として働く細胞であるリンパ球において、抗原受容体が機能を獲得あるいは拡張するために、その遺伝子が組換えを起こすことである。さらに、この組換えはリンパ球の分化そのものを規定している。リンパ球の抗原受容体の遺伝子は、可変部遺伝子の組換えによって部分断片から完成され、これによって個々の細胞がそれぞれ異なった抗原を特異的に認識・結合することが可能となり、免疫系全体として多様な抗原に対しそれぞれ特異的に反応する基礎を築いている。さらに、抗体はエフェクター分子として抗原の中和・除去などの機能も担うが、この機能を抗原の種類や侵入経路に応じて拡張するため、H鎖の定常部遺伝子が組換えられる。
本研究は、リンパ球の初期分化から最終活性化にたるまでにおける遺伝子組換えそのものとその標的遺伝子の特異性を制御する分子機構とそれぞれの特異性の鍵を握る因子(特に転写制御因子)は何かなどを解析することにより、組換え因子そのものの発現制御と、転写を介するこれらの組換えの標的遺伝子特異性調節機構の実態を解明することを目指した。その結果、クラススイッチ組換えの必須因子であるAIDの遺伝子発現調節、遺伝子組換えの標的特異性制御などに関与する因子としてPax5、 E2A、 SIP、 NFκB、 IRF4、 Runx1-3などが同定され、これらがTGFβ1シグナルの下流で作用するId2とともに、エピゲノム性調節制御の回路を形成していることを明らかできた。また、これらの因子群は、2次リンパ組織のダイナミックな構築変化と3次リンパ組織の形成による微小環境(場の制御)の影響を受け、時には冗長な、時には相反する機能を発現し、その制御を複雑化していることも判明した。加えてこればで培養条件下では効率よく誘導することが困難であったIgAへのクラススイッチ組換えを誘導する刺激の組み合わせを明らかにできた。
