研究概要

マウスの色素遺伝子slatyではメラニン生成が減少することが知られている。その機構を細胞培養系で調べた。Slaty(slt)マウスの培養表皮メラノサイトは、black(Slt/Slt)マウスのメラノサイトに比べ、第III期メラノソームが多く、成熟した第IV期メラノソームはきわめて少なかった。第III期メラノソームは、Slt/Sltマウスのメラノソーム(楕円形で内部構造が格子状)に比べ球形で内部に小顆粒を持った構造をしていた。また、Slt/SltマウスのメラノサイトはSlt/Sltマウスのメラノサイトに比ベゴルジ体やミトコンドリアが多かった。さらに、Slt/Sltマウスの表皮メラノサイトの増殖能はSlt/Sltと同等であったが、分化能が低かった。この培養系にメラニン生成の出発物質であるL-チロシンを加えて培養すると分化が促進され、ユーメラニンもフェオメラニンも増えた。Slt/Sltマウスでは、メラノサイトの分化に重要なチロシナーゼの活性やチロシナーゼ関連タンパク質2の発現も弱く、フェオメラニン量は変わらなかったものの、ユーメラニン量が少なかった。電子顕微鏡を使って調べると、L-チロシンは第IV期メラノソームの形成を促進し、楕円形で内部構造が格子状のメラノソームを増やすことがわかった。一方、recessive yellow(e/e)マウスのメラノサイトは増殖・分化活性が低いが、L-チロシンを加えて培養するも活性が上がった。e/eマウスでは雌の毛のユーメラニン・フェオメラニン量が雄に比べて多かった。e/eマウスの新生児に性ホルモンを投与してその影響を調べたところ、雄性ホルモンより、雌性ホルモンは少ない量でメラニン生成を促進した。以上のことから、recessive yellow遺伝子はMclr遺伝子と相互作用してユーメラニン生成を促進し、L-チロシンによって活性化され、発現が性ホルモンによって制御されていると示唆される。
