研究概要

個々のホスホイノシタイドに特異的かつ高親和的に結合するPHドメインをQ-dotプローブに結合させ、全く新しい考えに基づくホスホイノシタイドの微量定量、可視化を確立した。本法により、同時に3種のホスホイノシタイドを定量、可視化できた。FBP17やCIP4がF-Barドメインというホスホイノシタイド結合ドメインを持ち、膜変形作用があることを発見した。更に、それらが多量体フィラメントを形成して、細胞膜をチューブ状に変形させ、内向きの突起を形成し、エンドサイトーシスに関与している事を明らかにした。また全く新しいホスホイノシタイド結合ドメインとしてSH3YL1蛋白質のSYLFドメインを見つけ、マクロピノサイトーシスに関与している事を示した。
