研究概要

隅角線維柱帯組織の異常は緑内障を発症させる。隅角線維柱帯組織の構成成分であるヘパラン硫酸の役割を解明した。マウスでヘパラン硫酸が欠損すると神経堤細胞の分化が阻害され、緑内障を発症した。神経堤細胞の分化誘導におけるTGFβ_2シグナリングは、細胞表面に発現するヘパラン硫酸に依存していた。ヘパラン硫酸とTGFβ_2との相互作用を阻害する事で、線維芽細胞の活性化を抑制し、濾過手術の成績を動物眼で改善することができた。
