To evaluate the hypothesis that PRSb is the result, rather than the cause, of leaf flattening growth and to examine the relationship between PRSb expression and cell division activity, PRSb and KN1 expression will be examined for mold-treated J. prismatocarpus plants using in situ hybridization, and cell division will be monitored using EdU assay.

In the last year, the expression level of key genes including PRSb , KN1, and DL was checked, using quantitative RT-PCR, in both mold-treated samples and control samples of J. prismatocarpus. Preliminary results revealed that PRSb expression is reduced in some mold-treated samples as compared to control samples. Nonetheless, the expression level of these genes among samples fluctuated in a wide range (even for control samples). Therefore, additional experiments are being carried out. In addition, a round shaped species, J. wallichianus, was used as a second control, which should have low PRSb expression. Results of PRSb expression level of mold-treated samples of J. prismatocarpus will be interpreted together with control J. prismatocarpus (flat shape) and J. wallichianus (round shape). In addition, a few other mold designs were tried, with the aim to induce ectopic margins. This experiment is currently in process.
At the same time, the condition of non-invasively examining the shape of mold treated J. prismatocarpus leaf using X-ray computed tomography scan (CT) was established. This is helpful to know the leaf shape before using sample for qRT-PCR.
