In this fiscal year, I elucidated novel function of NADPH oxidase enzyme, Dual oxidase (Duox), in a pest insect Riptortus pedestris. Duox is originally well-known enzyme related to gut mucosal immunity of insects. Upon intestinal infection of pathogens, Duox generates bactericidal reactive oxygen species (ROS) and kill invaded pathogens. However, in R. pdestris Duox was not related to gut immunity, but it stabilized insect's respiratory network by cross-linking tyrosine bonds of proteins resulting in forming dityrosine network (DTN). Genetic blocking of duox gene indeed destabilized a DTN and it caused the collapse of insect respiratory network. Thus, aerobic symbiont Burkholderia were died from the midgut since they can't utilize enough oxygen. This study was published in PNAS journal.
