癌の進展におけるTGF-βシグナルの分子作用機構の解明

研究概要

TGF-βはEMTだけでなく、長期間の培養により筋線維芽細胞様細胞に変化するEMyoTも誘導することを見出した。その分子機構として、TGF-βがFGF受容体のスプライシングを変化させ、FGF-2に感受性を持つようになることがわかった。さらにFGF-2シグナルはTGF-βを協調的に増強し、EMTを非常に促進する一方で、TGF-βによるEMyoTを抑制することを見出した。その分子機構はTGF-βはMEK-Erkを抑制することでEMyoTを惹起し、FGF-2はMEK-Erk再活性化するために、EMyoTを抑制し、EMTを増強することがわかった。
