研究概要

癌抑制遺伝子CHFRは細胞周期M期チェックポイント機能を有し、E3ユビキチンリガーゼをコードするが、その標的基質分子および制御機構は不明な点が多い。本研究では、CHFR結合分子としてPARP-1を同定し、CHFRがこの標的分子である1本鎖切断DNA結合性のDNA 修復関連タンパクPARP-1のポリユビキチン化およびタンパク分解を介してM期チェックポイントを制御していることを突き止めた。またChfr欠損マウスおよびヒト口腔癌由来の細胞においてPARP-1タンパクレベルが有意に上昇していることを確認した。本研究はタキサン系抗癌剤抵抗性メカニズムの解明に貢献し、この抵抗性を示す口腔癌細胞において微小管阻害剤とPARP阻害剤とが有効な併用効果をもたらす分子機序を提唱する。
