摂食抑制ホルモンNesfatin1の脂肪分化抑制機構の解明と肥満治療への応用

研究概要

【目的】Nucleobindin-2(以下Nucb2)の脂肪細胞分化に対する作用を検討した。
【方法と結果】3T3-L1細胞を用いた。ウエスタンブロットによると、Nucb2の発現は脂肪細胞へ分化するに従い減少した。3T3-L1細胞のconditional mediumとヒトの血漿中にNucb2が存在することをウエスタンブロットで確認し、血漿中には凡そ50nMの濃度で存在すると計算された。種々の濃度のGST-Nucb2を3T3-L1細胞の培養液中に、脂肪細胞への分化開始48時間前から、24時間または48時間ごとに添加し、分化開始後8日目にoil red染色で脂肪細胞への分化の程度を評価した。その結果、50nMで明らかな抑制効果を得た。RT-PCRにてPPARγ、adipsinのmRNA量を、ウエスタンブロットにて発現量を評価したところ、同様の結果を得た。Nucb2からはNesfatin-1が形成されるので、GST-Nesfatin-1、GST-ΔNesfatin-1[Nucb2からNesfatin-1部分を除いたもの]について同様の検討を行ったが、脂肪細胞への分化抑制作用は認められなかった。
【考察】Nucb2には脂肪細胞への分化抑制作用が存在する。
