研究概要

生体内で一酸化窒素(NO)産生に依存して生成されるニトロ化 環状グアノシン一リン酸(8-ニトロcGMP)の心血管系における機能を検討した。フェニレフリン収縮血管に対し、8-ニトロcGMPは濃度依存性に弛緩反応と収縮増強を起こした。この収縮増強に対し、血管内皮由来のNO合成酵素アンカップリングで生じる活性酸素種の関与が示唆された。2型糖尿病マウス由来大動脈でのフェニレフリン収縮に対し、8-ニトロcGMP は濃度依存性弛緩のみ示した。以上より、内皮機能障害の改善に8-ニトロcGMPが寄与する可能性が示され、糖尿病合併症進展機構と8-ニトロcGMPとの関連性が示唆された。
