研究概要

近年,蛋白質をコードしないnon-coding RNAであるmiRNAが,蛋白質の翻訳抑制に働いていることが明らかとなり,post-transcriptionalな発現制御機構として注目を浴びている.miRNAは,mRNAと完全相補鎖を形成する場合にはmRNAの分解に働き,一部ミスマッチが認められる場合には翻訳抑制が起きる.miRNAは,発生や細胞分化の研究分野で精力的に解析が進んでおり,がん関連遺伝子の発現制御に関わるmiRNAも次々に特定されている.我々は,骨肉腫においてmiR-127の標的であるBCL6がそのfamilyであるNACC1の機能制御に関与している事を明らかにした.骨肉腫の発生・進展過程にはmiRNAの異常や蛋白の翻訳後修飾によるゲノム非依存性の機構が存在することが明らかとなった.
