研究概要

慢性骨髄性白血病(CML)の骨髄または末梢血CD34陽性細胞において、造血幹細胞や白血病幹細胞の自己複製に重要な役割を果たしているBMI-1の発現は慢性期、加速期、芽球期の順に高値であり、CMLにおける病勢進行に強く関与していることが明らかとなった。従ってBMI-1は病勢移行の分子マーカーの一つとして有用であり、この蛋白を分子標的治療の標的として考えうることが示唆された。次にリンパ腫ではBMI-1高発現細胞株はサーバイビンの発現亢進を介してエトポシド及びオキサリプラチンに対して耐性を示すことが明らかとなった。
