第VIII因子活性増強抗体を用いた血友病A新規抗体療法に関する基礎的研究

研究概要

これまでに、第VIII因子活性を増強する抗体の報告はない。抗第VIII因子モノクローナル抗体mab216は第VIII因子活性を1.5倍以上増強する抗体である。さらに、第X因子生成反応を1.5倍やトロンビン生成反応を2.5倍増強した。抗体の結合部位はA2ドメインと考えられ、本抗体の存在によりArg372の開裂が亢進していることが判明した。本抗体は血友病Aの止血治療に有用であることが示唆された。
