研究概要

p97は細胞内の様々な反応に関与している必須なAAA ATPase であり,優性進行性疾患である骨パジェット病と前頭側頭葉型認知症を伴う遺伝性封入体筋炎の原因因子である。p97には2つのAAAドメインがあるが,疾患を引き起こす変異はN末端側半分に集中している。本研究では,各ドメインのATPase活性を失った変異体を作製し,その機能を調べた。その結果,C末端側のAAAドメインはp97の機能に必須であり,その活性はN末端側のAAAドメインによって調節されていることが分かった。
