研究概要

病原性大腸菌O157は臨床的に最も注意を要する下痢原性の大腸菌である。O157の主要病原因子の一つは志賀毒素(Stx)であるが、Stxには遺伝的に異なる2種類の毒素が存在する。いずれのstx遺伝子もファージ上にコードされている。O157では、株間でStx産生量に大きな違いが存在することが知られているが、その理由は明確になっていない。本研究では、Stx産生量の違いに起因する遺伝的要因の解明を目指し、Stxファージの比較解析を行った。日本各地で分離された約50株のO157について、Stx1とStx2産生量を調べた結果、いずれのStxについても、株間で産生量にかなりのバリエーションが見られた。各株の系統解析を行ったが、株の系統と産生量の間には顕著な相関は見られなかった。Stx1ファージ3つとStx2ファージ5つの全ゲノム配列を決定して比較したところ、ファージ間でかなりのバリエーションがみられたことから、Stxファージの違いがStx産生量に関与している可能性が示唆された。現在、ファージのタイプと産生量との関係を詳細に解析しているところである。
