This research applied neurite orientation dispersion and density imaging, free-water imaging, and fixel-based analysis to baseline and follow-up data of subjects with Parkinson's disease (PD). As results of cross-sectional studies, we showed different patterns of brain tissue microstructural changes (i.e., neuronal loss and neuroinflammation) in different clinical subtypes of PD (i.e., with vs. without neurocognitive and psychiatric symptoms, tremor dominant vs. postural instability and gait disorder motor subtypes, with vs. without mild cognitive impairment, and with vs. without glucocerebrosidase gene mutations). Furthermore, in a longitudinal analysis, we demonstrated white matter neural compensation among PD patients with normal cognition one year after diagnosis.
