研究概要

マウスの生殖細胞の発生過程において、雄性生殖細胞の性分化の誘導にはNanos2の発現が必須であり、その誘導にはTGF-b（Nodal）シグナルを介したSmad2シグナル系が必要であることを明らかにした。Nanos2は生殖細胞の雌化を抑制し雄化を促進する。その際、Nanos2はRNA結合タンパク質Dnd1と直接結合して標的RNAを認識し、脱アデニル化活性をもつCNOT複合体と直接結合してRNAの分解に関与することが明らかになった。またNanos2は生後の精子幹細胞に発現し、GDNFを介した幹細胞の維持に必須である。Ｎanos2は主に分化抑制を介して幹細胞の維持に寄与する。
