研究概要

癌細胞表面に高発現するアミノペプチダーゼN(APN)を標的として新規低分子化合物を合成し、癌細胞の増殖や血管新生に対する抑制効果を検討した。新規合成されたヒドロキサム酸誘導体の一種である化合物24Fは、細胞表面に発現するアミノペプチダーゼの活性を阻害した。24Fを種々の肝癌細胞に対して作用させたところ、癌細胞の増殖及びコラーゲンゲルへの浸潤を抑制した。さらに、24Fを血管内皮細胞に対して作用させたところ、血管新生において重要な現象であるとされる細胞の移動や血管様構造の形成を阻害した。また、ベスタチン誘導体として合成された化合物LYPについても同様に検討を実施した結果、癌細胞の増殖を抑制するとともに、血管内皮細胞の移動や血管様構造の形成を阻害した。以上の結果から、本研究で新規に合成されたアミノペプチダーゼ阻害剤は、癌細胞の浸潤や腫瘍組織周囲の血管新生の抑制を介して肝癌の治療薬として有用であることが示唆された。
