研究概要

非小細胞肺癌の標的治療の感受性モデルを構築するために、肺癌細胞株パネルの感受性データベースを作成し、関連および感受性関連遺伝子のマクロアレイを用いたトランスクリトーム解析などの結果と抗癌剤感受性との関係を解析し、この結果に、薬剤の分子標的のシグナル情報解析(Pathway Architect software)を取り入れた。この解析により、HDAC阻害薬感受性を識別するsupport vector machine algorithm modelを構築した。加えて、HDAC阻害薬に影響される因子を解析し、それを基に併用治療開発を行なった。HDAC阻害薬とS-1の治療は有望な治療と考えられる。
また、PKC阻害薬について検討し、JAK/STAT signalling pathwayが感受性に関わり、実際にJAK阻害薬や、JAK高発現細胞で機能的に、その関与が証明された。我々の研究結果は、JAK1パスウェイがPKC阻害剤の感受性予測に有用であると考えられた。
