研究概要

各種膠原病患者を対象とし、PA28γ(Ki)およびPA28αのリコンビナント蛋白、および精製20Sプロテアソームを抗原源とした免疫プロット法にてプロテアソームに対する免疫応答を解析した。その結果、一連のプロテアソーム構成蛋白間およびそれらの結合蛋白に対する免疫応答の間には有意な相関を認めるものの、非結合蛋白戸の間には有意な相関が認められないことを明らかにした。また、プロテアソーム複合体の定量系を確立し、各種プロテアソームおよびその結合蛋白に対する抗体価が流血中プロテアソーム抗原量と相関する事を示した。以上の結果より、プロテアソームはその細胞内機能と関連しながらユビキチンを介して結合する一連の蛋白に対する免疫応答を誘導していることが示唆された。
