研究概要

本研究では、造血組織におけるTIF1bの役割を解析し、TIF1bが赤芽球の成熟、骨髄球細胞分化及びに造血幹細胞の骨髄再構築能に重要な役割を果たすことを見出した。さらに、網羅的発現解析やクロマチン免疫沈降法等によりTIF1bがp21遺伝子座に結合し転写抑制に働く事を明らかにした。また、TIF1b欠損細胞ではヘテロクロマチン蛋白質であるHP1aの蛋白質量が減少しており、HP1aがTIF1bによるp21の発現抑制に機能すると考えられる。以上の解析から本研究ではTIF1bによるp21の抑制が造血幹細胞機能に必須の役割を果たす事を明らかにした。
