研究開始時の研究の概要 |
We aim to investigate the hitherto unknown changes in physical and cognitive functions in the elderly from exposure to exercise, diet and brain training-based cocktail intervention, and also the concomitant alterations in plasma free amino acids and gut microbiota. The findings should help to establish a novel non-pharmalogical intervention aimed at preventing or improving aging-associated physical frailty and cognitive impairments, and might be crucial for countries like Japan with a super-aging society.

研究実績の概要 |
Three cohorts of elderly participants, ranging from 75 to 83 years old, have successfully concluded their 18-month interventions. These interventions encompassed a regimen of strength, balance, and endurance training coupled with cognitive function exercises or consumption of 100g of yogurt fortified with arginine and bifidobacterial LKM512 or adherence to their regular lifestyle. In our comprehensive analysis, we have meticulously examined demographic and clinical variables, as well as crucial biomarkers such as blood polyamines, amyloid-beta, and cognitive function metrics. In line with our findings, we are preparing detailed reports to elucidate their implications. Furthermore, we are actively performing the statistical analysis of additional variables, notably blood metabolomes.

理由
As planned, we have completed the quantitative measurements of plasma free amino acid concentrations in blood samples collected as different time points using the high-performance liquid chromatography-electrospray ionization-mass spectrometry. Also, shotgun metagenome sequencing of stool samples for gut microbiota collected at baseline and after intervention have been completed. In accordance with the hypotheses of our study, we are currently engaged in meticulous data analysis to discern potential changes in various variables from baseline to the 6th, 12th, and 18th months of intervention, as applicable. These data are undergoing statistical comparisons both within and between the groups, in line with our research objectives.

今後の研究の推進方策 |
We are in the process of drafting our initial manuscript, which will encapsulate the observed preliminary findings of this study. Subsequently, we will publish our research findings in esteemed, peer-reviewed journals, ensuring that our research contributes meaningfully to the scientific discourse and informs evidence-based practices. Notably, preliminary findings have already been presented at the BCSIR Congress-2023 in Dhaka, Bangladesh, underscoring the significance and potential impact of our research. Moreover, we are committed to disseminating further results at domestic and international conferences, fostering collaboration and knowledge exchange within the global research community.
