研究成果の概要

Activation-induced cytidine deaminase (AID) による抗体記憶のゲノムへの刻印はDNA切断により開始される。DNA構造の維持に働くトポイソメラーゼ1 (Top1) がAIDに誘導され、そのDNA切断を遂行する分子機構を解析し、従来のDNA編集仮説を正した。AIDはDNA切断とDNA修復という2つの機能を有し、各々はアミノ基側とカルボキシル基側の別々のドメインに依存する。各々の機能に特異的かつ必須のAIDの共役分子、hnRNP KとhnRNP Lの2つを同定し、それらの機能を解析した。AID突然変異と非常に似た病態の高IgM血症5型を示すウラシルDNAグリコシラーゼ (UNG) 欠損症ではCSRが抑制されるもののSHMは保たれており、2者の差を分子機構のレベルで証明した。また、APエンドヌクレアーゼ1 (APE1) のCSRでの機能がDNA切断後修復に特化していることを証明した。AIDによるDNA切断とDNA修復の2つはそれぞれ、特異的なクロマチン構造を要求する過程であることを証明した。クロマチン構造変換因子SMARCA4や転写伸長因子Spt4/5複合体、ヒストンアセチル化リジンに結合するBrd4が免疫グロブリン遺伝子組換えにおいて重要な機能を果たすことを示した。
