研究概要

スキルス胃癌は、高頻度に腹膜転移をきたす。癌幹細胞は、転移と関連している。本研究は、スキルス胃癌の病態を癌幹細胞の観点から解明し新規治療法を開発することを目的とした。スキルス胃癌幹細胞の転移能の解析、抗癌剤耐性能の解析、およびスキルス胃癌幹細胞とニッチ線維芽細胞との相互作用を解析した。その結果、スキルス胃癌には癌幹細胞が存在し、腹膜播種形成に重要で、CD44、integrinの関与が示唆された。胃癌幹細胞はUGT1A1が亢進することで抗癌剤耐性に関与していた。c-Met阻害剤は胃癌治療に有効であることが示唆された。さらに、ニッチ線維芽細胞はTGFβによってスキルス胃癌の幹細胞性を維持していた。
