日本住血吸虫宿主RNA取り込み関連遺伝子に関する研究

研究概要

住血吸虫はC. elegansと同様にlong-dsRNAのsoaking法によるRNAiが誘導できる。これまでの実験で住血吸虫は外界からdsRNAや、宿主のRNAを取込んでいることがわかってきた。このRNA取込みの意義を確認するために、RNAi関連遺伝子の一つであるsid-1遺伝子についての解析を行った。sid-1遺伝子は宿主に侵入してすぐの幼虫期で発現が上昇し、dsRNA刺激で発現が調節された。しかし、sid-1遺伝子ノックダウンにおいて、RNAi、dsRNA取込みや、寄生適応には影響を与えず、住血吸虫には独自のRNAi機構が備わっていると推測された。
