炎症性皮膚疾患におけるIL-33の免疫学的解析

研究概要

インターロイキン33(IL-33)はIL-18と同じIL-1ファミリーのサイトカインで、IL-18と同様にアトピーなどのアレルギーへの関与が指摘されている。そこでまず、ヒトならびにマウスの血中IL-33値のELISAを用いた測定系ならびに共焦点レーザー顕微鏡を用いたIL-33の免疫染色の実験系を確立した。IL-33は正常ケラチノサイトの核内に認められ、ブタクサによるアレルギー性結膜炎モデルマウスやハプテン頻回塗布による接触皮膚炎モデルマウスでは、IL-33が蛋白レベルで増加していた。さらに、DNFBをハプテンとする即時型接触過敏反応を検討したところ、野生型で観察される耳介腫脹がIL-33受容体ノックアウトマウスでは有意に低下していた。これらの結果は、IL-33が即時型接触過敏反応において重要であることを示唆している。
