研究開始時の研究の概要 |
In this research, a synaptic cleft-targeted copper ions-responsive labeling reagent would be developed to detect synaptic Cu release in the brain. The reagents would be synthesized first. The reagents would be evaluated in living cells to verify the proof of principle and assay the Cu-responsive properties. The optimal reagent would then be applied in cultured hippocampal neurons to detect synaptic Cu release in hippocampal neurons upon stimulation. Then LT-CuR would be used to detect synaptic Cu release in the more complicated biological environment, such as in acute brain slices and in vivo.

研究実績の概要 |
This year, I applied optimal CuR to ATP7A knock out cells and identified labeled proteins by mass spectrometry to study the misregulation of copper ions in living systems. I have also collected data to publish the results of this research, which I have submitted to a journal for review. Optimal CuR structure will be used to develop cuprous ion probes with organelle or synapse cleft localization.
