研究成果の概要

ポリケタイド合成酵素やクマロイルCoAリガーゼなど、天然物の生合成に関わる二次代謝酵素の中には、極めて寛容な基質特異性と潜在的触媒能力を有するものがあり、人為的な機能制御と分子多様性創出の格好のモデルとなりうる。本研究では、Ｘ線結晶構造解析を基盤として、タンパク工学の手法を駆使することにより、さらなる酵素触媒機能の拡張に成功した。また、人工基質を作用させることなどにより、一連の新規超天然型天然物を創出した。加えて、カリクリン、テレオシジン、アンチマイシンなど、一連の複雑骨格生物活性二次代謝産物の生合成遺伝子クラスターのクローニングと生合成マシナリーの詳細の解明に成功した。
