研究概要

マウスの発生過程で最初に造血幹細胞を認める胎生10.5日胚の大動脈-生殖原基-中腎領域において、造血幹細胞の発生に関わる血管内皮細胞に隣接する血液細胞塊の細胞に、転写因子Sox17および同じファミリータンパク質Sox7とSox18転写因子の発現を認めた。そこで、Sox17を血液細胞塊の細胞に強制発現すると、長期造血再建能をもつ造血幹細胞を含む血液細胞塊を in vitroで多数得ることが出来た。さらに、Sox17強制発現細胞よりSox17の発現を消失させると血液細胞への分化誘導が起きた。これらより、Sox17ファミリータンパク質はAGM領域における血液細胞塊の維持に必要であることを示した。
