研究概要

ティシュエンジニアリングの手法を用いた膵島細胞シートを開発した。in vitroにおいてインスリン分泌機能を持つ事、in vivoにおいて同系移植モデルで血糖値の正常化を確認している。今回は、同種移植モデルの系で、BALB/cマウスの膵島を単離し、温度応答性培養皿にて接着培養を行い、培養２日後に回収した細胞シートをB6マウスの皮下に移植後、血糖値の測定と組織学的検討を行った。血糖値の正常化は移植後３日目より認め、移植部位には炎症性細胞浸潤が確認された。マイトマイシン処理により約１週間の生着延長と細胞浸潤の抑制効果を認め、インスリンおよびグルカゴン陽性細胞から構成される細胞集塊が確認された。
