研究概要

肝実質細胞とクッパー細胞が共存する人工肝補助装置の開発には、クッパー細胞の多様な機能を解明することが重要である。IL-17Aノックアウトマウス(IL-17AKO)の検討からIL-17Aは肝細胞障害を防御するkey cytokineであった。またIL-17AKOとワイルドタイプマウス(WT)に肝切除を施行し、IL-17AKOでは肝再生が遅延することを明らかにした。またクッパー細胞を中心としたサイトカインカスケードに脾臓が関与していることを脾臓摘出モデルから検討した。その結果WTの脾臓摘出モデルでは肝再生が遅延した。肝再生には脾臓由来のCD４陽性リンパ球が産生するIL-17Aがきわめて重要である。
