研究開始時の研究の概要 |
This project will characterize the metabolomic profiles of phylogenetically diverse Cyanobacteria to identify links between exometabolomes and (1) their chemical diversity within the clade, (2) the organisms' phylogenies, and (3) nutrient limited growth.

研究実績の概要 |
The purpose of this project is to characterize cyanobacteria's exometabolomes through a combination of experimental and theoretical methodologies. At the beginning of FY2023, strains for the analyses were selected and have since continuously been cultured in optimal and nutrient limiting growth media. For the experimental part of this project, metabolomics analyses have been conducted for two strains and samples for two more strains are ready for analyses in both conditions. These data show the diversity of chemicals produced across strains and growth conditions. After receiving computational data at the beginning of 2024, it was integrated into a metabolic model for one strain for the theoretical work. Work with the model on this single strain is currently being conducted. As of now, media composition is reflected in the model and metabolomics data has been incorporated where possible. The data integration for other strains will be started once the computational protocols have been tested and fixed. This approach allows for the quantification of metabolites produced, including those metabolites that are produced inside the cell and those that are secreted but may have been below the detection limit for chemical analyses.

理由
Metabolomics samples have either been analyzed or prepared for analysis. Biomass composition samples have been prepared and protocols finalized, so that experiments can be carried out.
For the computational part, a pipeline for data integration and flux balance analysis is currently being established, so that once all wet lab data is available, analyses can be carried out for all strains.

今後の研究の推進方策 |
Metabolomics samples for almost all strains have been prepared and analyses for remaining strains will be completed until the fall. In addition, biomass compositional analyses for all strains will be conducted until the end of 2024.
Computationally, a pipeline for metabolomics data integration and flux balance analyses will be finalized by the fall. Biomass compositional data will then be added into the metabolic models before conducting flux balance analyses for all strains until the end of the fiscal year. Additionally, from the fall, phylogenomics studies including the analysed strains will be carried out.
