Modified the amino acids through plasma and understood the structure changes in proteins and peptides in the absence and presence of co-solvents. Additionally, to understand the effect of plasma-modified protein action on cancer cells, we will bind the protein with drugs and inhibit their action.

| 研究実績の概要 |
This research conducted during the last fiscal year resulted in four main outcomes. (1) Computational studies give an advantage in examining protein structures that are costly to analyze the biophysical properties following plasma treatment. Therefore, I studied the effect of plasma oxidation and electric field on the human mitochondrial heat shock protein (mHsp60) using molecular dynamic simulations. In addition to the possible plasma oxidation state, we also applied an electric field (0.003 and 2.0 V/nm) to check the changes in the Hsp60 protein. The simulation results show that the electric field strongly affects the mHsp60 structure compared with only oxidation state. The combination of oxidation and the electric field effect increases the destabilization of the mHsp60 structure compared with their respective control states. This is our citation as the Front cover of the Journal. (2) Another simulation's result published reveals that increasing nitration levels and stronger electric fields significantly accelerate pore formation in cell membranes. This phenomenon underscores nitration's modulatory role in altering electroporation dynamics. (3) In another experimental result, we employed DNA-labeled microbeads as biological indicators for the rapid visualization of plasma sterilization. (4) Further, we have shown the CO2 capture and conversion by the plasma treatment, that can play an important role in the anti-inflammatory action, anti-apoptotic effects, antiproliferative properties, or protective properties for tissues from hypoxia and reperfusion injury.

There is some delay in the computer simulation of the proteins, as the supercomputer was upgraded, which delayed its access for 4 months.

| 今後の研究の推進方策 |
We are further planning to test the effect of plasma treatment and electric field effect on more proteins computationally, as the experimental analysis is more costly, so before applying for another grant to check the effect of protein on the cancer cells, we will do so. As the experimental analyses are costly and need more budget. So this year I am planning to apply for another JSPS Kiban project for in-vitro analysis.
