研究成果の概要

本邦における家族性膵臓がんの原因遺伝子探索のため22家系47名を集積し全エクソン配列解析を行った。非同義性一塩基多様性、スプライス部多様性、翻訳領域内挿入欠失を選択し、1000 Genome、Human Genetic Variation Databaseで頻度1%未満の稀な多様性を抽出した。既知及びマウス遺伝子改変実験による膵臓がん関連遺伝子に着目して候補遺伝子を絞り込んだ。結果、BRCA2、PALB2、PUM1、FARP1、FAM193A、CTNNA1、MLL5に原因となる可能性のある一塩基多様性、挿入欠失を見出した。また、家族性膵臓がん及び関連腫瘍の臨床・分子病理学的特徴を明らかにした。
