研究概要

未分化型胃がん（DGC）は、発症数と死亡数が多い胃がんの中でも特に予後が悪い。我々はヒトDGCに形態学的にも分子生物学的にもよく似た胃がんを発症する世界初のマウスモデル（DCKO）を作製し、その胃がんから初代培養細胞株（MDGC）を樹立することにも成功した。
マイクロアレイ解析の結果より、DCKOマウスのDGCではエピジェネティクス変化が生じていることが明らかになった。そこで、MDGC細胞株にエピジェネティクス治療薬を処理し、スフェア形成を顕著に阻害する薬剤を複数同定した。それらの薬剤はin vivoでも腫瘍増殖抑制効果を示し、エピジェネティクス治療薬によるDGC治療の糸口が得られた。
