研究概要

アトピー性皮膚炎や高齢者では皮膚バリア機能低下により、難治性の皮膚炎を生じ、患者のQOLを著しく低下させる。本研究ではバリア機能の改善と抗炎症作用を合わせ持つ外用剤の開発を目的とした。まず、我々はコレステロールに注目し、実験系としてオキサゾロンによる遅延型皮膚炎モデルを用いた。結果、コレステロール外用群ではコントロール群に比べて有意に耳介腫脹反応が軽減した。また、コレステロールの外用群では細胞内コルチゾール活性化酵素である11β-HSD1の上昇がみられた。以上の結果より、コレステロールの外用は11β-HSD1の発現増加を介して、表皮の増殖抑制作用、炎症抑制作用をもつのではないかと考えられた。
