眼内薬物治療の飛躍的効果向上の研究：自然免疫の関与について

研究成果の概要

非感染性の眼内炎に焦点を当てて研究を行った。まずトリアムシノロン注射後に強い炎症を起こす病態について、結晶構造物による刺激が炎症を誘発すると考えて、培養R28細胞を用いて、トリアムシノロン顆粒がどのように影響を与えるかを調べた。その結果、結晶状の構造物を眼表面に持続的に接触させると濃度時間依存性に炎症性サイトカインが誘発された。次に細胞破砕物であるhistone H3を網膜色素上皮細胞に添加すると、炎症性サイトカインの産生が誘発された。過剰に添加するとアポトーシスを引き起こした。これらの作用にはいずれも自然免疫系で重要な働きをしているtoll like receptorが関与していた。
