研究概要

重症喘息を特徴付けるT細胞レベルのステロイド抵抗性のメカニズムを解析し、治療介入の前提となる前臨床研究を行う目的に、ステロイド抵抗性喘息モデルを樹立した。OVAで感作したマウスからT細胞を選択的に単離し、抗原刺激、限界希釈を行い、T細胞をクローニングした。in vitroにおけるT細胞クローンのステロイド感受性と、T細胞クローン移入マウスに抗原チャレンジを行うT細胞移入喘息モデルを用いて、in vivoでのステロイド感受性を評価した。さらに治療介入をめざして、costimulatory signal阻害薬、および、低分子量シグナル伝達阻害薬のステロイド抵抗性改善効果を確認した。
