喫煙関連頭頸部癌における浸潤能獲得、薬剤耐性獲得機序の解明と新治療法の開発

研究概要

本研究は、喫煙について、発がん要因のみならず、化学放射線治療の耐性化機序、浸潤能獲得機序の解明のため、頭頸部がんを含む細胞株に喫煙抽出物を曝露させた上で評価を行い、将来の喫煙関連頭頸部がん制圧を目的としているが、以下のごとくの研究成果であった。
１、頭頸部扁平上皮癌細胞株3種類について、喫煙曝露細胞株の抗がん剤、分子標的薬感受性を評価し、シスプラチン、EGFR阻害薬の耐性を確認した。また、肺癌、膀胱癌細胞にて浸潤能の新規獲得を確認した。２、次世代シークエンサーを用いた409がん抑制遺伝子のシークエンスにより、新規獲得変異を確認した。３、miRNAアレイにより、喫煙暴露関連miRNAを抽出した。
