In this study, we aim to develop a novel DDS for the targeted administration of zinc. A further objective is to elucidate the specific role of zinc in modulating parasympathetic innervation. During the first year of the project, we examined the effects of zinc on salivary gland development. Using an in vitro culture system, we found that zinc supplementation significantly enhanced glandular tissue growth and notably increased the elongation and distribution of neural cells. To validate these findings, we employed a mechanically stiffness-controlled culture environment designed to inhibit salivary gland tissue growth. Remarkably, the addition of zinc promoted tissue development. To explore the underlying mechanisms, we utilized various signaling pathway inhibitors. Our results indicate that zinc's influence on both salivary gland tissue growth and parasympathetic neural activity is mediated through the AKT signaling pathway. These findings provide novel insights into the critical role of zinc in the early developmental stages of salivary gland tissues and fulfilling one of the primary objectives of this study. Regarding the DDS, we have successfully synthesized the compound cyclo[RGD(DMNPB)fK], and future work will focus on evaluating its performance and further use in developmental processes with greater precision. In parallel, we worked on electrochemical sensor modifications, nano and microparticles synthesis. Using the obtained results, we have presented five papers at domestic and international conferences and published two academic papers.

理由
We have successfully presented significant research findings at the international conference, achieving one of the goals of this study: to elucidate the specific role and underlying mechanisms of zinc in the submandibular gland and the parasympathetic neural innervation. These results, derived from our details experiment, contribute valuable new insights to the field of life sciences. Overall, the research is proceeding smoothly and in line with the intended goals.

今後の研究の推進方策
In the future, we plan to examine the results obtained so far, particularly the functional ability of cyclo [RGD(DMNPB)fK] on salivary gland tissue growth, using cyclo[RGD(DMNPB)fK] and the RGD sequence alone. We will continue to optimize the material use as a method of local DDS. We also plan to use a similar method to investigate the effect of zinc on neuron cells. We also plan to conduct more detailed quantitative evaluations. As for in vivo, we aim to construct a gel sheet to use for localized zinc supplement, and will further proceed with qualitative and quantitative evaluations
