研究成果の概要

EGFR変異陽性ヒト肺腺がん株、HCC-827とPC-9細胞株にTGF-betaとFGF-2を曝露すると上皮間葉移行が誘導された。PC-9ではSmad3、HCC-827ではSmad3、MEK/Erk、mTOR経路の活性化を認めた。mTOR阻害剤（PP242）、メトフォルミン、DMSOはそれぞれ上皮間葉移行をリバースした。上皮間葉転換は化学療法抵抗性を惹起し、PD-L1発現を亢進させたがそのリバースにより、部分的に薬剤感受性を回復し、PD-L1発現を抑制した。Pirfenidoneとnintedanibも上皮間葉移行をリバースした。以上より上皮間葉移行は新たながん治療標的になると考えられた。
