In this study, we are trying to clarify the effects of prenatal exposure to perfluorinated chemicals on Glucocorticoid hormones in next generation. Among whole participants in Toho cohort (n=514), participants with available perinatal PFOS/PFOA levels in serum samples obtained in Sapporo Toho cohort were included in this study (n=295) after exclusion of twins and preterm births. Maternal PFOS and PFOA have been measured after the second trimester of pregnancy in serum samples by liquid chromatography-tandem mass spectrometry (LC-MS-MS). Baseline questionnaires, birth records and follow-up questionnaires were collected during pregnancy until 7 years old including information on parental demographic characteristics, dietary habits, socioeconomic status, birth size, children’s growth and infectious diseases (data cleaning has been performed). Genetic polymorphisms were assessed in last summer to determine of maternal (blood samples taken at delivery) and infant (cord blood samples) single nucleotide polymorphisms (SNPs) of glucocorticoid receptors. Genetic analysis were performed by EP1 system (Fluidigm). Cortisol and cortisone levels in cord blood samples of 130 randomly selected subjects have been measured by LC-MS-MS, and I started to do data analysis.
