研究概要

機能性蛋白質に対し、生理的条件下で構造をとらないが活性を持つ変異体や構造をとるが機能を持たない変異体を作り出した。活性を保持する要因、活性を失う要因を、リガンド誘導折り畳みや結晶構造解析により、揺らぎが分子認識を促進していることや揺らぎが活性を制御していることを示した。また揺らぎの制御には、蛋白質分子全体を取り囲む水和水の水素結合ネットワークの形成が必須であることが明らかになった。
