研究概要

本研究の目的は、次世代のフィジオーム・システムバイオロジーシミュレーション基盤を開発することにある。また理論研究と実証研究を有機的に実施し、心臓の電気活動の分子レベルから組織・臓器レベルまでの統合的な理解を得ることも目的としている。
昨年度に引き続き本年度も、多階層の生体モデル構築・シミュレーションが可能なソフトウェア(PhysioDesigner)の研究開発を進めた。特に細胞内シグナルパスウェイ・ネットワークのモデル構築・シミュレーションソフトウェアのデファクトスタンダードとなっているCellDesignerとの連動性を高め、またより親しみ易いユーザーインターフェースの研究開発を実施した。その成果として、PhysioDesigner第一版ソフトウェアを公開した。これによって、フィジオーム・システムバイオロジーシミュレーション基盤の研究環境が整い、多階層生体機能学の確立の礎ができた。
一方、不整脈の発症や治療に関連する実験研究も実施した。心筋細胞の電気的興奮性の制御で重要な役割を果たすhERGチャネルの研究では、開口促進作用を有する薬物の作用を系統的に解析し、結果として阻害とは異なる相互作用によってチャネルの活性化ゲート機能が変化すること、薬物毎にゲート機能への作用の強度が異なることを示した。さらに、この作用に関わる薬物のファーマコフォアモデルの構築に成功した。これにより、化学物質の構造からhERGチャネルに対する作用予測も可能になった。また、心拍のリズム制御に重要な役割を果しているG蛋白質制御カリウムイオンチャネル(Kir3.2)と無機イオンとの相互作用を解析し、チャネルの閉口機能や阻害の構造的基盤を明らかにした。これらのカリウムチャネルの分子制御機構の解明は、薬物や無機イオンの心臓機能への影響の理解と新たな抗不整脈薬の開発につながる。

理由

本年度の主たる目的であったPhysioDesignerの初期構築がほぼ完了し、統合環境プラットフォームソフトウェアとして第一版の公開が完了した。このリリースにより、領域内の研究者のみならず、領域外の研究者もこの基盤ソフトウェアの利用が可能となった。さらに、複数の心筋細胞モデルがデータベースに登録され、モデルデータベースが拡充された。

今後の研究の推進方策

現状、研究遂行上の問題点はなく、計画の変更の必要はない。今後も研究計画に沿って研究を実施する。特に、基盤ソフトウェアの操作性のさらなる向上を図り、各種データベースとの連携、その他の有用なソフトウェアとの連携をより強化する方針である。
