ゴキブリ類の社会進化に関する分子生態学的研究

研究概要

今年度は、昨年度から引き続いて、雌雄ペアとその若虫からなる明確な家族構造を持つクチキゴキブリ類を用いて、亜社会性を規定する遺伝子の探索を進めてきた。八重山諸島のタイワンクチキゴキブリを用い、分散前の成虫(有翅虫)とペアリング後の若虫と同居する成虫をそれぞれ雌雄に分け、キット(promega, RNAgents)を用いて頭部からトータルRNAを抽出した。その後cDNAを作成し、13コのプライマーセットでDifferential Display法を試した。いくつかの有翅虫特異的・成虫特異的なバンドを特定し、再増幅した後、大腸菌を用いてクローニングし、配列の決定を試みた。有翅虫雄特異的バンドから3つのシークエンス、有翅虫雌特異的バンドから2シークエンス、成虫雄特異的バンドから2シークエンス、成虫雌特異的バンドから2シークエンスの計9シークエンスを得た。全ての配列においてホモロジー検索を行ったが、特に高いスコアの配列は同定されず新規の遺伝子であると考察される。今後は、発現部位の同定など、各遺伝子のキャラクタライゼーションから機能の考察を行うことや、亜社会性行動の行動学的解析と対応させていくことが必要であると考える。
今年度は本プロジェクトの最終年度であるが、3年間の研究により、オオゴキブリ類の各分類群の系統関係に基づいた社会性や食性等の生態特性の進化パターンに関して極めて重要な知見を得ることが出来た。結果はすでに論文にまとめ、Proceedings of the Royal Society of London B誌に投稿し受理されている。
