コンディショナルノックアウト技術を駆使した熱ショック蛋白質の新機能解析

研究概要

70kDa熱ショック蛋白質(HSC70)は細胞質に豊富に存在し、種を越えて保存されている分子シャペロンで、細胞内蛋白質の品質管理に重要な役割を果たしている。
本研究では、細胞機能におけるHSC70の役割を解析する目的で、HSC70コンディショナルノックアウト細胞の作成を試みた。しかし、HSC70-like pseudogeneが多数存在したためターゲッティングが困難であった。そこでHSC70と結合し、その共役分子シャペロンであるHSP40ファミリー分子について遺伝子解析と機能解析を行い、そのターゲティングを試みている。データーベース検索により、新規HSP40ファミリー分子を3種類クローニングした。このうち2つの分子、DNAJ-B12とDNAJ-C8は細胞内でHSC70と結合し、いずれも細胞質及び核に局在することが判明した。組織発現では、前者は大脳、前立腺組織に多く発現し、後者は骨格筋、心筋、中枢神経に発現していた。DNAJ-C8に関しては、神経変性疾患であるポリグルタミン病の原因遺伝子の一つatrophin-1とのアミノ酸相同配列があり、現在抗体を作成してポリグルタミン病の病態形成への関与について解析中である。さらに、小胞体に局在する新規HSP40ファミリーを2種類クローニングした。そのうち、ERdj3に関しては上皮細胞や形質細胞に豊富に存在し、分泌蛋白の品質管理に関与していることが示唆された。現在siRNAを用いた遺伝子発現ターゲティングノックアウトを試みている。
一方、リンパ球の細胞表面抗原に対する抗体で、細胞死を誘導することのできる単クローン抗体Hal-1を樹立し、その抗原遺伝子の同定を試みた。レトロウイルスを用いた発現クローニングの結果、細胞表面にHal-1抗原を高発現する細胞株を樹立することに成功し、現在Hal-1遺伝子について解析中である。
