癌における糖鎖合成酵素の遺伝子発現と臨床応用

研究概要

1.マウスβ1ー4Gal転移酵素のcDNAを用いて、遺伝子DNAの単離に成功した。全長は65kbあり、7つのエクソンから成る。このうち、第3と6エクソンはalternative splicingを受けることが分かった。各エクソンの機能ドメインを決定した。調節領域と思われる5上流域はTATAやCAATボックスは含まず複数のGCボックスを有する。F9癌細胞を分化するレチノン酸が本酵素遺伝子を誘導する分子機序を検索している。ラット肝癌で亢進する2ー6シアル酸転移酵素(ST)のcDNAオリゴヌクレオチドを用いて肝癌のmRNAを調べたが増量はなかった。しかし、肝癌の本酵素から6kdのベプチドを切り出すプロテアーゼ活性は肝より肝癌が3倍高く、2ー6STは転写後の修飾によって活性が高まる可能性がある。2.癌で増量するGD_3の合成に働く2ー8STをラット肝ゴルジ画分から部分精製し諸性状を解析した。GM_2合成のβ1ー4GalNAc転移酵素をマウス肝から分画を行なう一方、GM_2に関するコンジェニックマウスからcDNAライブラリーを作り、GM_2^+とGM_2^-マウスのmRNAをプローブとして+・-法でクローンを探している。nLcOse_4に働く二つのラットGlcNAc転移酵素のうち、β1ー3酵素活性は脾Mn^<2+>要求性であるが、β1ー6酵素は二価陽イオンを必要とせず、肝癌では両者とも亢進していた。結腸癌のムチン糖蛋白合成に働くαGalNAc転移酵素はペプチドのーSerーThrーThrーに親和性があり、これらアミノ酸残基にGalNAcを高密度に結合する。このことは特異抗体で癌ムチンが高率に検出されることを裏づける。3.Le^x糖鎖合成に働くフコース転移酵素は、268例の癌患者血清で陽性率は66%、健常人7%であった。本酵素のレベルは癌の予後と並行しており、新しい癌マーカーとなり得る。硫脂質の合成酵素は腎細胞癌組織でも患者の血清でも亢進しており、腎細胞癌のマーカーとして臨床応用が可能である。
