肝化学発癌過程の前癌細胞における遺伝子および遺伝子発現の異常の解析

研究概要

佐藤はラット肝前癌病変の特異的マーカー酵素グルタチオンSートランスフェラーゼ胎盤型(GSTーP)や同じくヒト各種臓器癌マーカー、ヒト胎盤型(GSTーπ)の酵素的特性を、DNA組換え体GSTーPとその部位特異的変異体を大腸菌に発現させて解明し、47番目のシステインがSH修飾剤や活性酸素に高い感受性を有することを明らかにした。また村松とともにcーjun,cーfos癌遺伝子の発現がGSTーPの発現に深く関連することを明らかにした。村松はGSTーP遺伝子の5'上流にある強いエンハンサーGPEIにTRE(TPAーresponsive element)様配列が2つ回文状に並んだ特異な構造があり、cーjunが極めて低いF9細胞でもよく働くことを明らかにした。林はmyc癌遺伝子の癌細胞における発現機構として、myc mRNAの不安定配列を解明し、その安定化因子を究明した。またPCR法を利用したRNAの微量迅速定量法を確立し、極めて微量のRNAから特定のmRNAを定量することを可能にし、微小癌組織での癌遺伝子発現の検索を可能にした。渡部は肝化学発癌で重視されている脂質過酸化物の除去に従来の細胞質性のGSTの他に、ミクロゾーム性のGSTが重要であることを明らかにした。また佐藤とともに発癌剤硫酸エステル体を不活性化する新しいGST分子種を同定した。野口はピルビン酸キナーゼ(PK)アイソザイム遺伝子発現制御機構を究明し、ラット肝化学発癌過程で肝型の発現低下はトランス作用因子の減少で説明されるが、M_2型の発現増加はトランス作用因子の増加のほかクロマチン構造の変化が必要と示唆した。津田は肝発癌の進展に伴いG6Pase、ATPase、γーGTP、G6PD、GSTーP、AFP、PKM_2が同時に偏倚する頻度が高くなること、特にGSTーP、G6PDの有用性を指摘した。今後の問題としてはGSTーPなどの発現に癌遺伝子(cーjun、cーfos)の産物が関与していることが明らかになったので、より詳しい機構の解明が必要である。
